Abstract
热休克蛋白90(Heatshockprotein90,Hsp90)是热休克蛋白家族的重要一员。研究发现Hsp90的很多客户蛋白在肿瘤的发生与发展过程中具有重要的作用,Hsp90因此成为重要的抗肿瘤靶点之一。其抑制剂格尔德霉素(Geldanamycin,GA)的衍生物等二十多种化合物作为抗肿瘤药物已经进入临床实验,并展示出良好的应用前景。Hsp90抑制剂已成为抗肿瘤药物及其先导化合物的重要来源。 本论文以Hsp90为靶点,利用体外相互作用、分子对接和Hsp90的ATPase活性分析等方法,对来自海洋微生物和植物内生真菌等306个结构新颖、独特的天然化合物进行筛选,从中发现了与GA结构完全不同,...
