dc.description.abstract文昌鱼的器官构造和基因组成都较为简单,是研究胚胎发育机制以及脊椎动物器官起源的理想材料。然而,作为一种新兴的模式动物,目前文昌鱼研究领域的常用实验技术仍比较单一。显微注射技术是发育生物学研究中的一项重要实验技术,但是至今文昌鱼卵显微注射技术的应用还很不成熟。本文以白氏文昌鱼为材料,优化了文昌鱼显微注射步骤,用诱导产出的未受精卵进行显微注射,建立了一套高效的文昌鱼卵显微注射方法,并分别以KCl、β-actin启动子驱动的LacZ报告基因重组质粒以及Rfp报告基因重组质粒为注射溶液,通过统计注射后的受精率、胚胎发育时的出膜率、正常发育率以及胚胎表达LacZ和Rfp信号的阳性率对建立的显微注射技术进...
