dc.description.abstract第一部分:6-磷酸海藻糖磷酸酶(trehalose-6-phosphatephosphatase,TPP)是结核分枝杆菌生长的必需蛋白,可做为高通量筛选抗结核药物的潜在药靶。本部分成功制备了结核分枝杆菌的TPP(otsB2/Rv3372)蛋白。远紫外CD光谱实验表明TPP的二级结构由54.92%α-helix、8.61%β-sheet和36.47%coil构成,且结构稳定性较低(Cm=2.099±0.134M)。ITC实验发现,在Mg2+存在下,TPP可以与底物6-磷酸海藻糖以摩尔比1:1发生中等相互作用(Kd=39.52±1.78µM)。因为尚未得到TPP蛋白或片段的晶体,我们通...
