Generation of anti-dextran monoclonal antibody and development of immunonephelometry for quantitative detection of dextran

The purpose of the study was to generate a monoclonal antibody against dextran and develop a method of immunonephelometry for quantitative detection of dextran. Dextran is covalently linked to carrier protein bovine serum albumin. An anti-dextran monoclonal antibody was obtained by hybridoma technology, which showed titer higher than 10(5) in mice ascites and the characteristics of the monoclonal antibody was studied. Then an immunonephelometric assay for quantitative detection of dextran was established, which showed a detection limit of 0.35 NTUs (nephelometric turbidity units) with a sensitivity of 5.5 mu g/mL. Further studies confirmed high specificity, accuracy and stability of the approach. Findings suggest that the newly-developed method is reliable and useful to quantitatively detect dextran. It can be applied in sugar processing to examine dextran and monitor the quality of sugar products.
