Abstract
Mindin是一种外分泌的、高度保守的细胞外基质蛋白,并在先天性和适应性免疫中发挥重要作用。然而它对结直肠癌的发生发展过程中发挥的作用,及其上游调控机制仍未明确。 本论文的研究首先通过Elisa实验结果表明:在结直肠癌患者的血清中Mindin的表达水平较正常人明显下降且其下降水平在疾病早期表现更为显著。其次,为了探究Mindin的调控机制,我们通过生物信息学网站预测出可能的转录因子,并通过双荧光素酶报告实验、ChIPDNA及EMSA方法均验证了其中的早期生长因子Egr-1可在转录水平上调控Mindin的表达。再次,Egr-1在人类结肠癌细胞SW480和HCT15上调控Mindin的mRNA及...
