dc.description.abstract目的 探索肾癌A498细胞系在氧化应激状态下自噬发生的机制及其对细胞凋亡的影响,为探索治疗的新方法提供实验依据和理论基础。 方法 1. 应用不同浓度H2O2以及不同作用时间分别干预A498细胞,通过检测其CCK-8、ROS(活性氧簇)阳性细胞数量,确定构建A498细胞氧化应模型的H2O2最佳作用浓度及时间。选择200μM为H2O2处理细胞的最适浓度,分别作用A498细胞不同时间后,Westernblot检测氧化应激对自噬相关蛋白LC3Ⅱ和p62以及HIF-1α(低氧诱导因子1α)蛋白表达的变化。 2. 设计并合成一段针对人HIF-1α基因的shRNA,退火连接构建重组载体并酶切鉴定,将...
