dc.description.abstract本研究针对二维液相在复杂样品分离中的不足,结合新的混合模式色谱,构建基于SephadexLH-20×RPLC联用的新型二维色谱方法体系,并探索该系统在中药材分析和分离制备上的应用。 构建SephadexLH-20与反相色谱联用的在线分析系统,以含多个化合物的模式混合物作为研究对象,通过探究各化合物的液相保留行为证明了葡聚糖凝胶在甲醇/水洗脱模式下可兼具尺寸排阻、氢键吸附、分配三种分离机制。以红花作为研究对象,在确定溶剂洗脱模式的基础上考察第一维SephadexLH-20柱的柱长、流速等因素对分离效果的影响,确定分离参数;选择适合该二维系统的接口;将构建的SephadexLH-20×RPLC联...
