Abstract
NF-κB是细胞内重要的转录因子之一,其调控细胞内大量基因的表达,这些基因参与机体重要的生理过程,如细胞存活和增殖、细胞凋亡、细胞应激应答、炎症反应和免疫应答。许多刺激都可以活化NF-κB信号通路,大部分刺激引起NF-κB的活化均汇集到IKK复合体。IKK复合体磷酸化IκBα,导致IκBα被泛素化并最终通过26S蛋白酶体降解。IκBα的降解会导致NF-κB二聚体的释放,自由的NF-κB二聚体进入细胞核,在细胞核中经过一系列翻译后修饰,NF-κB二聚体会结合到相应靶基因的启动子或增强子的κB位点从而发挥转录调控作用。由于NF-κB调控的基因参与众多生理活动,NF-κB调控发生紊乱将导致多种疾病,...
