Abstract
炎症小体是一个由多种蛋白形成的功能性复合物,在固有免疫中有重要作用。在细胞炎症反应中,炎症小体可以激活caspase1的活性,进而剪切GSDMD诱发细胞焦亡,同时还能介导细胞因子IL-1β和IL-18的剪切成熟和分泌。细胞焦亡的失调和炎症小体的异常激活都会导致多种自身免疫病。细胞焦亡是新划分的一种程序性细胞死亡方式,最近GSDMD蛋白的发现是细胞焦亡研究的巨大进展之一。GSDMD蛋白在被caspase1/caspase11剪切后,会执行细胞焦亡。但是GSDMD执行细胞焦亡的具体功能机制还未有报道。本文中,我们通过体内体外实验验证了GSDMD蛋白的作用机制。研究结果显示,GSDMD-N端片段在细...
