Variability of Anther Culture and the Technology Strategies for Developing Pract ical Photoperiod Sensitive Male Sterile Rice
本研究以培矮 64S/ 890 2 S F2 代中选出的 11个典型的光敏核不育单株进行花培 ,获得了 2 2个花培单个愈伤分化苗 (指由单个愈伤组织分化的一丛苗 ,以下简称愈伤苗系 )为本研究的试验材料。通过远安 ,海南不同生态条件考察来源于同一愈伤苗系的育性变化。结果表明 ,同一 F2 单株来源的不同愈伤苗系之间、以及同一愈伤苗系的不同单株间 ,结实率变异范围和变异系数较大。同一愈伤苗系不同单株结实均值均以海南高于远安 ,说明多数光敏核不育单株 ,具有长日条件下育性稳定和短日条件下育性易转换特性。分子标记分析未能检测到不同于双亲的结构变异位点 ,并证实来源于同一愈伤苗系的不同单株变异 ,是由于花培过程中多个花粉粒同时诱导、分化而来的结果。本文还讨论了籼型光敏核不育水稻的选育技术路线The materials of this study were 11 typical PSGMS plants selected fr om F 2 generation of Peiai64S/8902S, and 22 callus regenerated plants (CRP) wer e obtained from anther culture. The fertility of these plants were investigated u nder the different ecology conditions of Yuan′an and Hainan. The major results a re as follows Wider variation range and Larger variation coefficient of seedsett i ng rate were found both among different callus lines from the same F 2 plant an d among different plants from the same callus line. The seedsetting rate of the same plant was higher in Hainan than that in Yuan′an. It shows that most PSGMS p lants have the characteristic of fertility stability under long day condition an d fertility reversibility under short day condition. RFLP analysis have not dete cted the genetic variation among the DH plants. It shows the CRP variation that come from same callus might be induced and differentiated by multiple pollens ef fects simullaneoasly.
