Comparative Study on Different Type Callus in Sweet Potato

不同激素浓度配比下，徐薯１８ 叶片外植体形成愈伤组织的类型和频率不同；在２，４－Ｄ 浓度为２ ｍ ｇ／Ｌ的ＭＳ培养基上获得３ 种愈伤组织（Ｅ型、ＮＥ型、Ｇ型），其色泽、质地、细胞的形态结构、分化途径显著不同，其过氧化物同功酶和酯酶同功酶酶谱也表现出显著特异性．The process of dedifferentiation of sweet potato could be greatly regulated by plant hormones. Three types of callus (type E, NE and G) have been induced from young leaves of Xushu 18 on MS medium supplemented with 2 mg/L 2,4 D. Although they were formed in the same culture condition, some differences were found among them in morphology, color, cell structure, differentiation way and isozyme. Type E was embryogenic callus with typical cell morphology and high division rate, which could form normal somatic embryos. Isozyme analysis of peroxidase and esterase showed that there were many differences in isozyme spectra patterns among type E, type NE and type G, while little divergence was discovered between the patterns of type E cells and somatic embryo cells. This means that the isozyme spectra of peroxidase and esterase may be considered as physiological markers for distinguishing embryonic callus from non embryonic callus.
