Genetic Diversity of Castanopsis hystrix by RAPD Analysis

采用从100个随机引物中筛选出的30个有效引物,利用RAPD技术对不同地区的10个不同生态型优质红锥种源进行遗传多样性研究.结果表明:(1)30个有效引物共扩增出290条清晰谱带,平均每个引物扩增出9.67条带.多态性条带为254条,比率为87.59%;(2)10个不同生态型红锥之间的遗传相似系数介于0.594 6~0.714 8之间,但不同地区红锥也存在着一定的遗传差异(遗传差异在0.285 2~0.405 4);(3)用引物S3扩增出的带谱可在一次实验中区别10种不同生态地区的优质红锥,同时S3也可作为10种优质红锥材料的指纹图谱,鉴定优质红锥的种源;(4)聚类分析表明,10种优质红锥种源可聚成3个独立的类群.本文作者对它们的亲缘关系进行了推测.Ten assession of Castanopsis hystrix from different ecological areas was analyzed using the RAPD method,and 30 effective primers were selected from 100 random amplified primers.The result showed:(1) There are 290 amplified bands by 30 effective primers,with an average of approximately 10 bands per primer.Among the amplified bands,there are 254 polymorphic bands,percentage of polymorphic bands being 87.59%.(2) Genetic similarity index of ten C.hystrix from different ecological areas ranged from 0.594 6 to 0.714 8,and their genetic distance ranged from 0.285 2 to 0.405 4.Among the ten C.hystrix,there are obvious genetic diversity.(3) The ten assession of C.hystrix can be distinguished using the molecular fingerprint with the primer S3 in simple experiment,and the advantage quality of C.hystrix can be appraisted by using the amplified bands with the primer S3 also.(4)The ten assession of C.hystrix can be classified into three independent groups based on the polymorphism.There are obvious differences and abundant genetic variations among the ten C.hystrix assession.Their genetic reletive and genetic distance were discussed.
