The Purification of Chondromyces sp.

[中文文摘]从武夷山的一枯枝上分离得到了一株稀有软骨霉状菌,在培养过程中发现,该株黏细菌在人工培养基上只有在与一种未知的细菌共培养的状态下才能长出典型的子实体并保持菌种的活力.然而要从共培养的菌落中纯化这株黏细菌,使用常规的黏细菌纯化技术几乎是不可能.在这种情况下对伴生细菌进行16S rDNA菌种鉴定,再根据鉴定为施氏假单胞菌(Pseudomonas stutzeri)的生态特性和两种菌之间耐受性和营养需求方面的差异,采用有别于黏细菌传统纯化方法的水浴灭菌、酸性抑制和贫瘠营养等多种方法,成功地将伴生细菌除去,获得了该株软骨霉状菌的纯培养,这些方法为普通黏细菌的分离纯化方法提供了新的启示.[英文文摘]The Chondromyces sp. was isolated from a piece of rotting wood collected in Wuyi Mountains in August 2008. In the process of purification, we found that Chondromyces sp. could not live and produce fruit ing bodies unless cultivated together with an unknown bacteria. Once t he two species grew together, the unknown bacteria could be purified easily but not the Chondromyces sp.by traditional methods. Later, the unknown bateria was identified as Pseudomonas stutzeri using 16S r DNA sequense identification. It belongs to a kind of bacteria which cannot grow in low pH , high temperature and a poor culture medium, wereas the myxobacteria can grow . Finally, the Chondromyces sp. and the bacteria were separated by using a water-bath, an acerbic and a poor culture medium. This method broadened the way of t he purification of myxobacteria.
