Determination of andrographolide and dehydroandrographolide in Andrographis paniculata Ness by micellar electrokinetic capillary chromatography

Abstract
Andrographic paniculata nees is one of well-known traditional chinese medicinal materials (TCMs), in which andrographolide and dehydroandrographolide are two major active compounds. In this work, a micellar electrokinetic capillary chromatographic (MECC) method has been developed for separation and quantitative analysis of these two compounds in andrographis paniculata ness. Electroretic conditions are as follows: uncoated fused silica capillary (42.2 cm/50 cm (effective/total length), 75 mum i.d.), 20 mmol/L phosphate-10 mmol/L sodium dodecyl sulfate containing 5% methanol (V/V) at pH 6.8, applied voltage 20 W, temperature 25 degreesC, detection wavelength 240 nm, injection 25kPa.s. Both of the analytes were separated successfully within 10 minutes under the optimum conditions. The relative peak areas of the two analytes increase linearly with the increase of their concentrations in the range of 10.4 similar to 93.5 mg/L for andrographolide and 10.3 similar to 92.7 mg/L for dehydroandrographolide, and the revoveries are 102% and 102% respectively.
