dc.description.abstract通过测定黑曲霉β-葡萄糖苷酶的底物特异性,表明该酶不仅能水解纤维二糖和对硝基苯-β-d葡萄糖苷,还能微弱地水解对硝基苯-β-d-半乳糖苷和β-d-木糖苷。Ag-+、Cu-(2+)和Hg-(2+)对该酶有较强的抑制作用。该酶水解对硝基苯-β-d-葡萄糖苷、水杨苷和纤维二糖的kM值分别为2.32、19.11和30.18MMOl/l,V_(MAX)则分别为412、237和198μMOl·MIn-(-1)·Mg-(-1),lInEWEAVEr-burk作图法表明,d-葡萄糖和δ-葡萄糖酸内酯对该酶显示竞争性抑制作用,其kI分别为5.17和1.31MMOl/l。
dc.description.abstractSubstrate speciFicities and inhibitors of the puriFied β-glucosidase From Aspergillus niger were investigated.The relative activity of β-glucosidase toward p-nitrophenyl β-D-galactoside were 100,91.3,9.7 and 7.2.The Km values For P-nitrophenyl β-D-glucoside,salicin and cellublose were 2.32,19.11 and 30.18mmol/L.The Vmax were 412,237 and 198umol.min-1 mg-1.The enzyme activity was strongly inhibited by Ag+,Cu2+,Hg2+,D-glucose and D-glucono-δ-Lactone.The K1 For D-glucose and D-glucono-δ-Lactone were about 5.17 and 1.31mmol/L respectively,as determined by Lineweaver-Burk plot.
